Are the grids adjustable in both directions? From the picture it only appears that the vertical grids can be adjusted. Also how loose are they?

No they are not they are only removable horizontally to make the grids longer vertically. The grid dividers are very tight in there so no issues staying or holding things I place.

Are these storage boxes stackable?

I only have one but it looks like they can be stacked. It doesn't have anything that would hold it in that position though like groves or anything.

Can I use this as an advent calendar?

I suppose you could use this container as an Advent Calendar. I hadn't thought of it that way. I use mine to store various sewing machine accessories.
