My office has a cement floor that gets very cold in the winter. Will this mat hold up under a desk with a rolling office chair all day?

The high/low surface pattern of this particular mat would make it very difficult to roll around using an office chair.  You may want to check out the product below which can handle desk chairs rolling on top all day and still stay in place well on a cement (or any other hard) floor.

Yes

The Americans with Disabilities Act (ADA) addresses the use of mats in public places, where entrances and exits must be kept free of hazards.

Section 302.1
"Floor and ground surfaces shall be stable, firm, and slip resistant"

Section 302.2
"Carpet or carpet tile shall be securely attached...Exposed edges of carpet shall be fastened to floor surfaces and shall have trim on the entire length of the exposed edge."

"Pile height shall be 1/2 inch maximum."

Section 302.3
"Changes in level between 1/4 inch high minimum and 1/2 inch high maximum shall be beveled with a slope not steeper than 1:2."

Will it work on a bedroom carpet (protection from elderly incontinence) with a wheelchair?

These mats, by default, come with a universal gripper backing to grab the floor better, especially on carpeted surfaces.  The mats have a polypropylene surface (which is more stain-, fade- and mildew-resistant compared to cotton, nylon or wool) and dries faster as well.  The edges have a water dam border to prevent liquids from running off the sides.  The edges are also beveled so as not to be a tripping hazard and allow for wheels to roll on and off the mat more easily.  To clean the mats, you can shake out or vacuum (for day-to-day dirt and debris) and hose down (with a little dish soap as needed) or even power wash for a more thorough cleaning.
