How much sand do I put in this filter ?

Calls for 130 lbs. I bought three 50lb bags and used them all. I suspect I have more than needed and could have gotten away with just 100. I have checked the sand after running the filter for some time and all of the visible debris is captured within the top inch of the sand bed. Adding more than suggested just reduces the headspace above the sand bed which needs to be open for debris accumulation.

Have a 3500 gallon water Koi Fish pond.....will this system be ok ?

This system is more than adequate for your needs. A koi pond is a lot dirtier than a pool is. The extra size will prevent you from having to backwash frequently.

Where should the clear hose sit in the filter

That is an air relief tubing. It just needs to stand next to the center pipe and the end above the sand bed
