Because it’s a wide shelf, (10”), I’d use 4.

It depends on the weight you plan to put on the glass shelf. In your case, I suggest four. And there are 8 brackets in each product, which can 100% meet your needs:-)

5 is fine. And it is recommended to use toughened glass.

Q: What is the measurement from back of the bracket to the front of the bracket? How many mm or inches of the bracket is on the shelf?

They are about 1”. And 3/4 of the glass shelf is in the bracket. Really secure. I used a glass shelf with 2 brackets and put about 3 pounds on shelf with no issues.
